
So, on a mouse and keyboard, I play "FPS style" -- WASD are my movement keys, with A and D set to strafe instead of turn. I turn using click-drag with the right mouse button. As such, my "fastest" keys are the ones I can quickly reach with my left hand from the WASD cluster, and so I focus my binds to make those my most used keys. If you click abilities to activate them, your mileage may vary.
With that in mind, here's my setup:
For targeting allies, I use the F keys and a focus target. F1-F8 targets the appropriate party member, and I have F bound to "target focus target" and Shift-F bound to "set focus target". Unless I need it for something else, my focus target is basically always the tank. When healing, that makes it easy to keep him up, and when DPSing, it helps me target his target. An example of when I'd focus something else is a boss fight where I'm tanking or DPSing, where knowing what the tank is on is far less important than being able to monitor and quickly get back on the boss.
In the 8 person instances, I've also used focus target to keep an eye on a healer when I'm tanking.
For targeting enemies, I have T bound to "target of target", E bound to "target next enemy (nearest to farthest)", and Shift-E bound to "target previous enemy (nearest to farthest)". If the tank isn't marking, then, I use F to target him, then T to target his target. If he is marking, I'll use E and Shift-E to cycle targets to the correctly marked one. As Melkire notes, the target next/previous binds won't target anything you can't see, so you can limit the number of targets you have to cycle through by rotating your camera. Of course, for me, when there's enough targets that that's an issue, I'm usually AEing, and so then I'm targeting using E to find whatever mob in the melee has the highest health to be my AE target.
